    Modelling vibratory roller–soil system dynamics using discrete element method

    A simple yet useful implementation of lumped-parameter models using the discrete element method is discussed in this paper. Lumped-parameter models are often used in the modelling of vibratory roller-soil systems owing to their ability to capture the essential features observed during roller-soil vibrations. In order to investigate the system behaviour in the event of a decoupling (loss of contact between the drum and the soil), two conditions pertaining to the transition between the contact and non-contact phases are compared in the light of simulation results. The results demonstrate significant differences in the roller-soil system behaviour based on operating conditions. One particular case provides an unrealistic estimate of the contact force, and thus may lead to an incorrect prediction of the system dynamics. Another solution, however, provides a reasonable estimate of the system dynamics and is recommended herein

    Greenhouse Gas Emissions from Asphalt Pavement Construction: A Case Study in China.

    In China, the construction of asphalt pavement has a significant impact on the environment, and energy use and greenhouse gas (GHG) emissions from asphalt pavement construction have been receiving increasing attention in recent years. At present, there is no universal criterion for the evaluation of GHG emissions in asphalt pavement construction. This paper proposes to define the system boundaries for GHG emissions from asphalt pavement by using a process-based life cycle assessment method. A method for evaluating GHG emissions from asphalt pavement construction is suggested. The paper reports a case study of GHG emissions from a typical asphalt pavement construction project in China. The results show that the greenhouse gas emissions from the mixture mixing phase are the highest, and account for about 54% of the total amount. The second highest GHG emission phase is the production of raw materials. For GHG emissions of cement stabilized base/subbase, the production of raw materials emits the most, about 98%. The GHG emission for cement production alone is about 92%. The results indicate that any measures to reduce GHG emissions from asphalt pavement construction should be focused on the raw materials manufacturing stage. If the raw materials production phase is excluded, the measures to reduce GHG emissions should be aimed at the mixture mixing phase

    The Greenhouse Gas Emission from Portland Cement Concrete Pavement Construction in China.

    This study proposes an inventory analysis method to evaluate the greenhouse gas (GHG) emissions from Portland cement concrete pavement construction, based on a case project in the west of China. The concrete pavement construction process was divided into three phases, namely raw material production, concrete manufacture and pavement onsite construction. The GHG emissions of the three phases are analyzed by a life cycle inventory method. The CO₂e is used to indicate the GHG emissions. The results show that for 1 km Portland cement concrete pavement construction, the total CO₂e is 8215.31 tons. Based on the evaluation results, the CO₂e of the raw material production phase is 7617.27 tons, accounting for 92.7% of the total GHG emissions; the CO₂e of the concrete manufacture phase is 598,033.10 kg, accounting for 7.2% of the total GHG emissions. Lastly, the CO₂e of the pavement onsite construction phase is 8396.59 kg, accounting for only 0.1% of the total GHG emissions. The main greenhouse gas is CO₂ in each phase, which accounts for more than 98% of total emissions. N₂O and CH₄ emissions are relatively insignificant

    Effect of warm mix agent on the chemo-mechanical performance of binder with different oil sources

    In this study, three typical oil source asphalt binders, Karamay asphalt A, CNOOC 36-1 asphalt B, Qinhuangdao CNPC asphalt C, were selected to prepare asphalt binders together with the warm mix agent Evotherm M1. The effects of warm mix agents on asphalt from different oil sources were experimentally studied via dynamic shear rheological (DSR), thermogravimetric analysis (TG), and Raman spectroscopy tests. The asphalt binders with different oil sources exhibit different properties. The rheological test results indicate that the addition of warm mix agent can slow down the decrease of asphalt viscosity during the aging process and the aging of asphalt. The results of the thermogravimetric test showed that the residual mass of asphalt with the addition of a warm mix agent significantly decreased after aging. Warm mixing agents can slow down the conversion of lightweight components to heavy components during the aging process of asphalt. By calculating the reflectivity of asphalt in Raman spectroscopy, it can be concluded that the reflectivity of asphalt decreases after adding a warm mix agent. The warm mixing agent reduces the degree of thermal evolution of asphalt. Warm mixing agents can make the chemical components in asphalt relatively stable and less prone to further pyrolysis or cracking reactions

    Portland Cement Hydration Behavior at Low Temperatures: Views from Calculation and Experimental Study

    Environmental condition affects the property of construction materials. This study gives an initial understanding of Portland cement hydration under low temperatures from the views of laboratory experiments (including electrical resistivity, degree of hydration (DoH), and maturity) as well as thermodynamic calculation. The hydrates of Portland cement at the given period were detected with X-ray diffraction (XRD), and their microstructure was observed by scanning electron microscope (SEM). Experiment result (i.e., DoH and electrical resistivity) indicated that the hydration of Portland cement was delayed by low temperature without hydration stopping at −5°C. Based on a basic kinetics model, the thermodynamic calculation predicted that the final hydrate differs in dependence on environmental temperatures. The mechanical behavior trend of Portland cement paste affected by low temperatures potentially is linked to the appearing of aluminate compounds and reduction of portlandite

    Neutralization of Diverse Human Cytomegalovirus Strains Conferred by Antibodies Targeting Viral gH/gL/pUL128-131 Pentameric Complex

    Human cytomegalovirus (HCMV) is the leading cause of congenital viral infection, and developing a prophylactic vaccine is of high priority to public health. We recently reported a replication-defective human cytomegalovirus with restored pentameric complex glycoprotein H (gH)/gL/pUL128-131 for prevention of congenital HCMV infection. While the quantity of vaccine-induced antibody responses can be measured in a viral neutralization assay, assessing the quality of such responses, including the ability of vaccine-induced antibodies to cross-neutralize the field strains of HCMV, remains a challenge. In this study, with a panel of neutralizing antibodies from three healthy human donors with natural HCMV infection or a vaccinated animal, we mapped eight sites on the dominant virus-neutralizing antigen-the pentameric complex of glycoprotein H (gH), gL, and pUL128, pUL130, and pUL131. By evaluating the site-specific antibodies in vaccine immune sera, we demonstrated that vaccination elicited functional antiviral antibodies to multiple neutralizing sites in rhesus macaques, with quality attributes comparable to those of CMV hyperimmune globulin. Furthermore, these immune sera showed antiviral activities against a panel of genetically distinct HCMV clinical isolates. These results highlighted the importance of understanding the quality of vaccine-induced antibody responses, which includes not only the neutralizing potency in key cell types but also the ability to protect against the genetically diverse field strains. IMPORTANCE HCMV is the leading cause of congenital viral infection, and development of a preventive vaccine is a high public health priority. To understand the strain coverage of vaccine-induced immune responses in comparison with natural immunity, we used a panel of broadly neutralizing antibodies to identify the immunogenic sites of a dominant viral antigen-the pentameric complex. We further demonstrated that following vaccination of a replication-defective virus with the restored pentameric complex, rhesus macaques can develop broadly neutralizing antibodies targeting multiple immunogenic sites of the pentameric complex. Such analyses of site-specific antibody responses are imperative to our assessment of the quality of vaccine-induced immunity in clinical studies

    Amplification Refractory Mutation System (ARMS)-PCR for waxy sorghum authentication with single-nucleotide resolution

    Waxy sorghum has greater economic value than wild sorghum in relation to their use in food processing and the brewing industry. Thus, the authentication of the waxy sorghum species is an important issue. Herein, a rapid and sensitive Authentication Amplification Refractory Mutation System-PCR (aARMS-PCR) method was employed to identify sorghum species via its ability to resolve single-nucleotide in genes. As a proof of concept, we chose a species of waxy sorghum containing the wxc mutation which is abundantly used in liquor brewing. The aARMS-PCR can distinguish non-wxc sorghum from wxc sorghum to guarantee identification of specific waxy sorghum species. It allowed to detect as low as 1% non-wxc sorghum in sorghum mixtures, which ar one of the most sensitive tools for food authentication. Due to its ability for resolving genes with single-nucleotide resolution and high sensitivity, aARMS-PCR may have wider applicability in monitoring food adulteration, offering a rapid food authenticity verification in the control of adulteration

    Cycling comfort on asphalt pavement: Influence of the pavement-tyre interface on vibration

    Attainment of cycling comfort on urban roads encourages travelers to use bicycles more often, which has social and environment benefits such as to reduce congestion, air pollution and carbon emissions. Cycling vibration is responsible for the cyclists’ perception of (dis)comfort. How asphalt pavement's surface characteristics relate to cycling comfort, however, remains undiscovered. In this study, the cycling vibration intensity on 46 sections of 24 urban roads was tested using a dynamic cycling comfort measure system while the cyclists’ perception of vibration was identified via questionnaires; the cycling comfort was then defined based on the cycling vibration. To record the accurate pavement-tyre interface under a stable environment, a total of 19 pavement sections were scanned using a 3D digital camera. These 3D models were then 3D printed, which are used to conduct the pressure film test using a self-developed pavement-tyre interface test system. Three ranges of pressure films were adopted to characterize the pavement-tyre interface via 9 parameters, namely contact area (A c ), unit bearing area (B u ), stress intensity (S i ), stress uniformity (S u ), kurtosis (S ku ), spacing (Sp a ), maximum peak spacing (Sp max ), radius ratio (R r ) and fractal dimension (F d ), in consideration of the area characteristics, pressure amplitude, peak spacing and shape of the interface. Finally, the significant interface parameters were identified, and the regression model between interface parameters and cycling comfort was established. Results show that the cycling vibration was described to be ‘very comfortable’ when the human exposure to vibration level (a wv ) was less than 1.78 m/s 2 ; ‘comfortable’ when the a wv was between 1.78 m/s 2 and 2.20 m/s 2 ; and ‘uncomfortable’ when the a wv was greater than 2.20 m/s 2 . The average stress on rear wheel-pavement interface is higher than that of the front wheel. B u-0.6 , Sp a-0.6 , and F d-0.6 are significant to cycling vibration. The 2LW pressure film is recommended for use to measure the bicycle pavement-tyre interface. The recommended interface characteristics are less than 7 mm 2 of the unit bearing area, 6 mm of average spacing and 2.38 of fractal dimension. Finally, dense asphalt mixture performs better in providing cycling comfort than the gap-graded asphalt mixture. Results of this study contribute to current knowledge on bike lane comfort and pavement design, the findings should be interested in cyclists, transport planners, and road authorities

    From hyper- to hypoinsulinemia and diabetes: effect of KCNH6 on insulin secretion

    Glucose-stimulated insulin secretion from islet β cells is mediated by K channels. However, the role of non-K K channels in insulin secretion is largely unknown. Here, we show that a non-K K channel, KCNH6, plays a key role in insulin secretion and glucose hemostasis in humans and mice. KCNH6 p.P235L heterozygous mutation co-separated with diabetes in a four-generation pedigree. Kcnh6 knockout (KO) or Kcnh6 p.P235L knockin (KI) mice had a phenotype characterized by changing from hypoglycemia with hyperinsulinemia to hyperglycemia with insulin deficiency. Islets from the young KO mice had increased intracellular calcium concentration and increased insulin secretion. However, islets from the adult KO mice not only had increased intracellular calcium levels but also had remarkable ER stress and apoptosis, associated with loss of β cell mass and decreased insulin secretion. Therefore, dysfunction of KCNH6 causes overstimulation of insulin secretion in the short term and β cell failure in the long term.Yang et al. show that KCNH6 plays a key role in insulin secretion and glucose hemostasis in humans and mice. Dysfunction of KCNH6 results in a hyperinsulinemia phenotype in the short term and hypoinsulinemia and diabetes in the long term
